Research areas
- Melanocortin MC4R/MC3R agonist — acts centrally, not vascularly
- Studied for HSDD (hypoactive sexual desire disorder) in preclinical research
- Works independently of the PDE5 pathway — different mechanism than Viagra-class compounds
- Investigated in both male and female sexual response research
- Rapid onset in preclinical models — minutes to hours
- Lyophilized powder, 99% HPLC verified purity — COA included

PT-141 (bremelanotide) is a synthetic cyclic peptide activating melanocortin receptors, approved as Vyleesi for premenopausal women with acquired, generalized hypoactive sexual desire disorder (HSDD). It works through the central nervous system, not by altering blood flow.
| Dose | Frequency | Duration | Route | |
|---|---|---|---|---|
| Vyleesi Label Protocol (FDA-approved) | 1.75 mg | 1 dose per 24 hours (max), 8 doses per month (max) | On demand | Subcutaneous (abdomen or thigh) |
| Phase 3 RECONNECT trials | 1.75 mg | On demand | 24 weeks | Subcutaneous |
| Long-term safety extension | 1.75 mg | On demand | Additional 52 weeks | Subcutaneous |
| Phase 2 intranasal (men with ED) | 7.5-20 mg | In trial arms studied | Not specified | Intranasal |
Cycle: FDA-approved on-demand (Vyleesi) is 1.75 mg subQ ≥45 min before activity, ≤1/24h, ≤8/month. Phase 3 trials used 1.75 mg subQ on demand for 24 weeks, with a long-term extension for an additional 52 weeks.
Benefits and research observations: Pharmacodynamic effects begin 30-60 minutes post-dose, with a reported peak effect window of 1-3 hours. Nausea may start within 0-30 minutes and typically resolves within hours as effects taper over 4-12 hours.
